When mousing / hovering a thread title (preview) what setting is it to enlarge the text and line spacing

There's no stylevars for it. So you'll need to add custom CSS:

.b-topicpreview .b-topicpreview__previewtext {
    font-size: 16px;
    line-height: 1.4em;
}

Whenever I post custom CSS, it should go to css_additional template in AdminCP or CSS Editor in Sitebuilder. For vBCloud, the latter is the only option.

For self-hosted vB, both 2 options are available. For vBCloud, only the second one is available as editing templates is not allowed.
